Polymerase chain reaction (PCR) is an enzymatic DNA amplification process that selectively amplifies a particular segment of DNA in vitro. PCR is very sensitive and specific as it can amplify a single DNA molecule to millions of DNA molecules in a very short time. It can selectively amplify a particular segment of DNA even in the presence of DNA from other organisms.

Conventional PCR is an end-point analysis method, where agarose gel electrophoresis is required to determine the size of the PCR product.

Real-time PCR is the advanced version of conventional PCR. Real-time PCR allows both the enzymatic PCR process itself and detection of the PCR product to occur concurrently in the same reaction vessel. Real-time PCR uses fluorescence dye (SYBR Green I, TaqMan probe, etc). Measurement of fluorescence at every PCR cycler allows the product accumulation to be displayed on a plotted curve. The amount of fluorescence is proportional to the amount of PCR product in the reaction vessel. If a specific target DNA is present in a sample, the time or number of cycles it takes will generate a detectable signal. Use of this information of cycle number enables easy target DNA detection.
